چکیده

As estimated from initial uptake rates, the uptake of glucosamine-l-14C by cultured Novikoff rat hepatoma cells in glucose-free basal medium follows normal Michaelis-Menten kinetics, with an apparent Km of about 20 mM. Since glucosamine is not accumulated by the cells against a concentration gradient, the main mode of uptake is by facilitated diffusion, but other evidence indicates that glucosamine also enters the cells by simple diffusion. Glucosamine is probably transported by the same system as glucose. Glucosamine transport is competitively inhibited by glucose and, to about the same extent as is glucose transport, by Persantine and Cytochalasin B. Since neither of the two inhibitors affects the phosphorylation of glucose or glucosamine by cell-free preparations, we suggest that transport is a reaction distinct from phosphorylation. At low concentrations of glucosamine in the medium (0.1 mM and below), the inhibition of glucosamine transport into the cell by glucose, Persantine, or Cytochalasin B results in a proportional decrease in glucosamine incorporation into macromolecules. When cells are incubated with low concentrations of glucosamine-14C, most of the acid-soluble

HEPATOMA McARDLE-RH7777 CELLS HAVE THE SAME RESPONSE AS NORMAL RAT HEPATOCYTES TO BOTH DIBUTYRYL-cAMP AND ANTICALMODULINW-7

The effects of cAMP-analogue dibutyryl-cAMP and anticalmodulin W-7 were studied on de novo synthesis and secretion of lipids in cultures of hepatoma McArdle RH7777 cells and normal rat hepatocytes. Dibutyryl-cAMP and W -7 separately caused a significant decrease in the secretion of de novo synthesized triacyl [3H]glycerol in both cultures of McArdle cells and rat hepatocytes. The inhibito...

Metabolism and cytotoxicity of 5-azacytidine in cultured Novikoff rat hepatoma and P388 mouse leukemia cells and their enhancement by preincubation with pyrazofurin.

5-Azacytidine transport into cells was measured in the absence of metabolism in adenosine triphosphate-depleted and uridine kinase-deficient Novikoff cells. Azacytidine is transported with about the same efficiency as uridine and cytidine by the facilitated nucleoside transport system of these cells. The phosphorylation of azacytidine in untreated, wild-type cells, however, is much more inhibit...

Nucleotide Pools in Novikoff Rat Hepatoma Cells Growing in Suspension Culture

This study was undertaken to measure the absolute levels of nucleoside pools in Novikoff rat hepatoma cells (subline N1S1-67) during growth in suspension culture in the presence of high concentrations of various nucleosides in the medium, and to obtain further evidence for the compartmentalization of the nucleotides in independent cytoplasmic and nuclear pools. The levels of nucleotide pools we...

Inhibition of de novo pyrimidine nucleotide and DNA synthesis and growth of cultured Novikoff rat hepatoma cells and other cell lines by pyrazofurin (NSC 143095).

Pyrazofurin (PYF), a C-riboside, inhibited the replication of cultured Novikoff rat hepatoma cells, HeLa cells, and mouse L-cells at concentrations as low as 0.1 to 10 muM, but Novikoff cells were more sensitive than the cells of the other two cell lines. Inhibition of cell replication was completely prevented by the presence of 0.1 to 1 mM uridine in the medium, and partly by the presence of o...
